An innervated skin 3D in vitro model for dermatological research

Abstract A 3D in vitro model of innervated skin would be a useful tool dermatological research to study the effect different chemicals and compounds on sensory properties skin. Current models are limited composition often composed ex vivo explants and/or animal-derived material. In this study, our aim was develop human with better biomimicry for research. Fibrin hydrogel aligned electrospun fibers poly(ethylene oxide terephthalate)/poly(butylene terephthalate) (PEOT/PBT) were used as scaffold generate model. The component made primary keratinocytes fibroblasts, while neuronal iPSC-derived neurons. Our results showed that dermal consisted fibroblasts synthesized collagen. epidermal characterized by expression keratins 10 14, involucrin. Finally, neurons extended axons throughout reached epidermis. Treating capsaicin solution 30 min, which performed proof concept test sensitization studies, resulted into partial depletion substance P tubulin β3. This could studying skin-neuron interactions cutaneous toxicity.
